Players can choose to play as one of the three species in a variety of game types, including deathmatch and capture the flag. In addition to the single-player mode, Aliens Versus Predator 2 includes a multiplayer mode which supports up to sixteen players. Players must complete a series of objectives in order to progress through the storyline, using weapons and items specific to each race. The Predator campaign follows a clan of Predators as they seek a rare Xenomorph queen, while the human campaign follows a human marine sent to investigate the planet. The Alien campaign follows the species' attempt to regain control of a human-colonized planet.

The single-player mode of Aliens Versus Predator 2 follows three separate campaigns, one for each race. It is the sequel to Aliens Versus Predator, released in 1999. The game is based on the Alien and Predator franchises and follows a three-way war between the species.
